We need help as to how to word a "letter" with our save the date card to let prople know that they need to call to reserve their room as soon as possible. We do have rooms on hold, but the hotel would like people to call asap. The idea for the response card with the save the date card for an idea of how many people may attend was a great idea. Thank You. Many of the responses have really helped us already!

If it's not too many people, I would just call for a chat and then remind them.

If it's a lot and you really need a form letter, I would write something like:

In order to be sure that you will have the accomodations you will need to attend, we have reserved a block of rooms in (Name of Hotel). The management at (Name of Hotel) has requested that you call to confirm a reservation at your earliest convenience. Below is some information that should be helpful in choosing accomodations.
